Re-sterilization help

okay after a month of waiting i finally got my spores. upon the advice of another user he said to re-sterilize all my substrate. jars have no sign of growth anywhere on them. and have been kept in a pretty sterile environment.

so should i add some water to the jars when i PC them again?

also how should i clean my syringes they came loose(not in a bag). should i just wipe them down with ISO?

also are there any good techniques to inoculating popcorn or in general?
 
I can't say for certain about the jars as I've never grown mushies, but the syringe can be sterilized with some IPA.
Worst case with the substrate is you have to toss it and redo the sterilization.

don't reuse your substrate.


Sterilize your needles by wrapping them in foil, put the foil in a jar, don't let it touch the sides or the bottom of your Pressure Cooker.
 
maybw i should clarify, substrate was made and sterilized 3-4 weeks ago, unused, untouched in glove box. should i re sterilize? the popcorn has no growth as should be expected with properly steriized substrate.

also there are spores in these syriges :/ they came loose packed between some paper in the shipping box

It it was sealed, sure, use it. If not then don't, even if you think it is still sterile, you will have lost some moisture - it isn't worth the trouble you might cause.


I never have flamed my needles - it tends to discolor them and take away the temper.
 
so what are you saying use the jars as is? let me look at the moisture it think its about the safe from when they got put in jars honestly, my vent ports are really small. as far as sterile... all these jars got over an hour in the cooker lids are on tight and ive wiped them down once a week with ISO so they are pretty sterile i am confidant there at little risk of contam.
 
fire them up then, no resterilization - I've used petri dishes that have sat in a glove box for up to three weeks - the only problem is their moisture life is limited.
 
i think what im gonna do is PC most of them again and add 10cc of water to qt jars and 5cc to the half qt jars i think i will inoculate 2 of my current jars just to see if they are still good to use after this much time. PCing them should not be a problem i dont think... well i dont see any reason to go all out and make new substrate when this popcorn is probably fine.
 
you know what your probably right. im gonna take one last look at them here later once im all cleaned up and if i can perceive some moisture, and they sound moist not like little rocks ill go ahead and fire them up without PCing again.
 
If the inside is uncontaminated, all you have to sterilize is the top of the jar - you can do that in 15 or 20 minutes at 15 lbs

yeah i was thinking a short time if i do PC, if i add the water i will boil it and add it in with a syringe.
 
just wanted to thank you canndo for all your help and patience. its been 4 days since i took your advice and just inoculated the jars without any further treatment. got mycelium from 2 jars so far, both of my strains (B+ and GT) :) super happy!

What are u using for a substrate if it's something easy to get chuck the old refill jars and pc I wouldn't risk a print or syringe on something that I had to question wether would work or not .
 
Oops speed reading must have skipped ur last post hope they colonize good for u

its cool, i didnt really have doubts about the substrate i just needed a go ahead to be sure it was still good. my vent ports are small so gas exchange is limited so thier was little loss of moisture. everything seems good thus far tho.
