so i made an lc from spore syringe for the first time i always make them from clone tissue. anyways i checked on it after three or four days n there apears to be some mycelium growth possibly grouped around very very tiny black dots.. maybe spores or spore clusters?

so thats my question is it early contam or is it spores.. it does have the white cloud look that most of my tissue clones had just the tiny black dots are throing me off.

the small dots are about the size of spores you'd just barely be able to see in a syringe. not like a huge mass. i only used like .5ml at the most to innoculate my lc.

You can't see single spores with the naked eye. This is another reason one should work from agar. The only way to tell is to wait some more, or grow some of it out - unless someone here has a more ingenious method.

I understand that which is why i mention clusters. because you can clearly see the spore clusters in a syringe and in the syringe i used there where lots of small clusters after shaking it up... good ole sporeworks fills them to the brim.

Now i have no ability to use agar, so its completely off the table for now.

see the only thing throwing me off are the small dark dots. just trying to see if any one else was ever able to see something like that when making lc from spores. ive made quite a few lcs from tissue, just never spores, im only making it from spores because i wanted to prolong my very last few cc's of golden teachers as long as i can before buying more spores or making another spore syringe.

what if i pull some out an look at it under mini microscope? would i be able to tell from just like 30-100x magnification. I belive i have some slides still. would I be able to tell difference from the different spore stages? monokaryotic mycelium, or if somatagamy had taken place, dikaryotic mycelium, apart from being some type of contamination?

I have a feeling its contam.
 
I don't think your microscope is powerful enough to see what you need to see - squirt some in some sterile grain, just a drop or so and see what grows.
 
I think I know what you are talking about I had the same thing with my first lc. Sporeworks syringes come very heavy in one big glob that settles. Not like homemade ones where you get that nice distribution of "purple water." Kind of curious as to Sporeworks method of making syringes that causes the spores to bind together like that. Even after shaking the hell out of them there is still large clusters of purple floating around. I've only ever used sporeworks, but I've had friends order from other vendors and thought they got ripped off since there was nothing at all visible in the syringe. Anyway, when I made the lc, one of those small globs got into the lc. The mycelium mass forms around it until I had a cloud with a darkish center. Didn't hurt anything, although the glob is almost impossible to suck back when you go use some lc, so I keep away from it and try to pull from the outside. I put a marble in the jar beforehand to try to break things up, but it doesn't work as well as I had hoped. Next time I am going to pick up a magnetic stir rod for like $3. I have an old PC fan I am going to try to make a "stir plate" with.

How exactly do you make you lc from tissue? Take a core with the needle and just shoot it into the lc like you would spores?
 
Tissue clones can be done quite a few ways, i normally pick a nice big cap and clean it with peroxide then split the cap open and dig out some flesh from inside the cap with a knife or something just drop that right into my lc, I dont use the airtight silicon lids or anything just regular holes in the lid like it was a pf jar. you can also use a needle filled with sterile water and stab through the stem or carefully dig out some flesh from the cap and squirt it right into the jar. basically you just take some clean flesh from a mushroom and stick it in an lc it grows the cloudy mycelium super fast. best to pick a big mushroom thats also part of a cluster, gives better flushes.
