    I would keep it at the lowest setting where you still have negative pressure in the tent, and temperature and humidity stay in a good range. Any faster is not really necessary.

    I dunno, I have a slightly smaller tent than you and air gets pulled in the bottom right and exhausted out the top left and back into the room, and I've not run into any issues with that (in a concrete basement no less). I guess it might depend on how big the room is and how much it's sealed...

You can probably remove that few inches of ducting and just butt the filter right up against the fan and seal it as best you can...

    You could also try decreasing your air exchange by eliminating the active intake and/or decreasing the exhaust fan speed. Or exhausting back into the lung room instead of outside. Both of those things would help conserve some of the water the plants are transpiring instead of sending it outside...
