I have soaked cubes for 24 hours and for a few seconds and never had a problem either way. For me it has never made a difference that I could see but I still usually soak em overnight just to be sure.

Its hard to say, but anytime you change things around the plants can get super stressed. I would dilute the solution by 50% and make things as stress free as you can for em. No fan directly on em, light not too close, perfect temp in room...etc.
