So I figured I would start a thread for whatever is getting your Mad Scientist Rock's off? What's your latest Experiment's? Something people can have input on seemingly impossible endeavor's or just share their experience's in there science experiment.....Whatever it is your doing share it it does not matter if it's something new or not:

I used Premium Silicone Glue it's got a temp rating of 400 degree's to seal my hole I made with a nail in jar cap

I used 91% ISO Propyl Alc. Had Paper towel soaked in it to wipe port off and use to cool and further sterilize tip of needle...

Heated my needle(Red hot where I plan on Breaking the integrity of the jar), Cooled(Iso) then injected

Never tried making my own or even looked it up ...Kick the cook's ass LMFAO:clap:bongsmilie...That does sound cool actually, maybe I'll research that

hybridization is a deep topic, and very hard. It's not possible to cross strains of the same species, as a hybrid is a cross between two different species within the same genus, not two different strains. The only thing that is possible with two strains of the same species would be genetic mutations. For example albino penis envy was created by creating a homozygous mutation of PF albino + PE. this is a difficult process and probably not something I would even attempt, honestly.

Not to say that I couldn't, its just very tedious work. Although I have considered a simple approach to breeding mutations. It's just a theory at this point ;)

There are 3 different types of genetic mutations. Dominant, incomplete dominant (or sometimes co-dominant pretty much the same), and recessive.

When using the word mutation its easy to imagine something like PE. Mutation in this sense means mostly changes in color, not really shape (though I am sure there are more mutations like PE to be discovered).

Dominant mutations are not common with mushrooms. One incomplete dominant mutation that we know of is leucism (albino a+). An example of recessive mutations in cubes is PE and Albino PF. PE can revert back to a normal mushroom, which means that it lost a copy of the mutation on a DNA allele, and became heterozygous. This is how we know it is recessive, as it is an unusal type.

This is a very basic and unorganized description of genetics. If you are already familiar with this, then you already know ;)

So lets say we wanted to create a leucistic version of penis envy. This would be possible by isolating one spore of both specimens, the PE, and the Albino A+, then pairing them together until we saw dikaryotic myelium. The resulting mushrooms will all be heterozygous for leucistic penis envy. These mushrooms will not be white, nor will they look like PE at all. They will generally look like normal cubes. However, they are halfway there. All we have to do at this point is take spore prints from two fruits on the same flush, and repeat the procedure of obtaining a single spore from both prints and pairing both spores until dikaryotic mycelium has formed. The resulting fruits will be white PE with purple spores.

So you can see how it might be extremely hard to isolate two spores from a kagillion that is on the spore print. We need an easier way. My theory is not exactly precise. Its more like kicking in a door and spraying a room with a shotgun loaded with buckshot, hoping we hit something. But I think it will work. I'll get more into the actual process at another time when I actually attempt it.

They don't have a viscous type solution available maybe? Assuming your using a big clean room....Seem's like you got the hookups!!:p That would maybe help in keeping your spore's isolated and more immobile under a scope? Would something like Methyl Cellulose get the job done easier or would it damage the spore's?

Right on, Man. I did a liquid culture of some cubes a while back. Still have a couple spore prints that I made. Hrmmm, is it shroom season?

Gettin a little late in the season my way......But all we have anyway's are are trashy P. Sub...Mid Aug to Mid Sep around me depend's on the weather...Comin up on Oyster season though ain't it....
