I do it all the time. I usually harvest the top cola first, as it tends to ripen first, and then let the lower buds continue to go until they too are ready. It's very standard practice.

I start my seedlings in peat moss, then transfer to the HB. I would recommend you grow your mom in soil (much easier and stable) and take cuttings and clone in HB. That's what I do. Good luck.
